Important Update to OmniStain Protocols

We recently replaced methanol with tBuOH to better preserve endogenous fluorescence. However, some users have reported tissue distortion in large samples—an issue we had not previously observed. We are conducting a rigorous evaluation to determine which tissue types may be most susceptible and to identify the root cause.

In the meantime, to ensure reliable results, we are reverting this modification and recommend using methanol as the solvent for both tissues and organoids.

Additional adjustments for consistency:

• Room temperature: 20–25°C. As some users in cooler environments (e.g. parts of Europe) experienced variability.
• Process timing: Dehydration/rehydration steps have been extended from 15 minutes to 30 minutes each and frequency of dehydration/rehydration in 50% v/v methanol/H2O increased from once to twice to accommodate inter-operator variability.
